At Sistemic, we leverage the instructive power of microRNA (miRNA) to provide a broad range of leading-edge products for the Bioprocessing, Cell (and Stem Cell) Therapy, and Drug Discovery & Development markets. From robust quality control (QC) for cells and stem cells, to toxicity and drug response monitoring for drug discovery & development programmes. These products are helping companies globally to reduce the risk during development and deliver safe & efficacious therapies to the market.

The Sistemic Methodology

Sistemic’s pioneering products are focused around the company’s IP and core expertise within three areas, constituting The Sistemic Methodology:

  • Robust miRNA Profiling: miRNAs are seen as the master controllers of a cell and, as such, Sistemic believes they provide information on the effects within a whole cell or organism rather than an isolated pathway. This systems approach means that the data produced is more indicative of the biology and more informative of actual effect. As a result, we leverage miRNA based profiling/fingerprinting to provide robust and information rich products to companies worldwide.
  • Superior Statistical Analytics: We have developed proprietary statistical algorithms that robustly and with a high degree of confidence analyse changes to miRNAs when a cell is perturbed by a stimulus. Together with our innovative visualisation techniques, we are able to generate a fingerprint of a key set of miRNAs known as “key miRNAs” (kmiRs) that represent these changes accurately.
  • Multi-Layered Contextual Analysis: Our products not only provide a set of kmiRs, representing the change, but can also provide a detailed contextual analysis for those changes linking the kmiRs back to the biology for enhanced understanding.

The Sistemic methodology provides a rigorous yet rapid understanding of the biological changes that occur within a cell. Resulting data, allows companies to make well-informed decisions, in a timely fashion. This enables them to develop their cell (or stem cell) therapy and drug development pipelines more successfully and in a cost-effective manner.

Our Process

We work very closely with companies to provide them with solutions that will work for them and resolve their issues. We consider ourselves as value partner and work very hard to develop products of the highest quality. As a result we use ‘best-in-class’ technology at each step of our process to ensure that we deliver results of the highest quality. To meet the varying needs of our customers we have put in place a flexible yet rigorous process:

Total RNA Preparation

RNA suitable for miRNA analysis can be isolated from a variety of source materials: all cell types (human, animal), frozen tissue, paraffin-embedded tissue and body-fluids including blood. Sistemic can prepare total RNA in-house, or inform on the best methods of preparation using their rigorous in-house standing operating procedures (SOPs).

RNA QC

RNA samples are processed using NanoDrop and Agilent Bioanalyser 2100 to quantify and ensure integrity before proceeding to the profiling stage.

miRNA Profiling

We use a variety of microarray technologies, such as Agilent’s microRNA microarray, to profile miRNA. This is further validated by performing QPCR using Quanta BioSciences reagent to ensure robust profiling.

Statistical Analysis

A multi-dimensional approach is used to analyse the miRNA profile data. We apply a number of statistical analyses and visualisation techniques on the data to identify the key set of miRNAs significantly affecting the system.

Validation

Once the kmiRs have been identified, changes in expression can be confirmed by performing QPCR using the Quanta BioSciences reagent.

Contextual Analysis

With access to gene ontology databases, we can provide contextual analysis on the kmiRs identified by SistemRNA™, to help inform on the biological context of the data.
